Description

Description: This assay employs a two-site sandwich ELISA to quantitate YWHAH in samples. An antibody specific for YWHAH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any YWHAH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for YWHAH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of YWHAH bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: YWHAh product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse, rat and bovine orthologs. This gene contains a 7 bp repeat sequence in its 5' UTR, and changes in the number of this repeat has been associated with early-onset schizophrenia.The 1,730-nucleotide sequence of the cDNA contained a 191-bp 5-prime noncoding region, the complete 738-bp coding region, and an 801-bp 3-prime noncoding region with 3 canonical polyadenylation signals.
